Bilan de la toxicité aiguë
Le tribénuron-méthyl est faiblement toxique quelle que soit la voie d'exposition. Il est peu irritant pour la peau mais modérément pour les yeux et il n'est pas un sensibilisant cutané.

Le tribénuron-méthyl n'a pas démontré d'activité génotoxique ou mutagène dans les essais in vitro et in vivo (essais de Ames, mutations ponctuelles sur des cellules ovariennes de hamster chinois, synthèse non programmée de l'ADN sur les hépatocytes de rats, formation de micronoyaux dans la moelle osseuse des souris, etc.).

Une faible activité œstrogénique a été observée chez les rats femelles. Le tribénuron-méthyl et ses métabolites pourraient être des agonistes pour le récepteur œstrogène. Dans l'étude de cancérogénicité, le dérèglement hormonal s'est produit à une dose excessive.

Dans une étude sur la reproduction des rats d'une durée de 2 générations, la progéniture n'a pas démontré de sensibilité accrue comparativement aux parents. Aucun effet sur la fertilité n'a été observé, ni sur le nombre de naissances, ni sur la survie.

Dans une étude sur le développement des rats et des lapins, les fœtus n'ont pas démontré de sensibilité accrue comparativement aux mères. La toxicité observée chez les fœtus était causé par une toxicité maternelle.

L'impact du tribénuron-méthyl sur le système nerveux n'a pas été spécifiquement évalué. Cependant, aucune évidence de neurotoxicité ou de neuropathologie n'a été notée dans les études aiguës, subchroniques, chroniques ou sur la reproduction et il n'y a pas d'inqui`tude concernant la neurotoxicité du développement. Bilan des effets à long terme
Le tribénuron-méthyl n'est pas mutagène mais il a été classé cancérogène possible pour l'humain en raison de l'augmentation significative d'adénocarcinomes des glandes mammaires chez les rats femelles. Cependant, cette augmentation due possiblement à un dérèglement hormonal s'est produite à une dose excédant la dose maximale tolérée et il est peu probable que cet effet survienne à un taux plus bas. À une dose plus faible, une baisse de poids corporel a été observé chez les rats. Dans une étude chronique chez les souris, on a observé une dégénérescence bilatérale des tubes séminifères et une oligospermie alors que dans une étude d'un an chez les chiens, des changement à la biochimie clinique ont été notés. Une étude subchronique chez les rats a démontré, entre autres, une diminution du poids corporel, de la consommation de nourriture et du poids absolu de certains organes. Le tribénuron-méthyl n'était pas toxique pour la reproduction et le développement dans les études animales. Il n'est ni génotoxique ni neurotoxique.
